Descripción de la Microbiota Intratumoral y su Asociación con la Respuesta al Tratamiento de Cáncer Colorrectal según Sexo: Un Análisis Secundario Exploratorio de Datos Metagenómicos

Abstract

El cáncer colorrectal (CCR) es una de las principales causas de morbilidad y mortalidad a nivel mundial. En los últimos años, la microbiota intratumoral ha sido propuesta como un factor relevante en la progresión tumoral y en la respuesta al tratamiento. Asimismo, el sexo biológico podría influir en la composición y función del microbioma, lo que plantea la necesidad de estudiar estas interacciones en conjunto. El objetivo del presente estudio fue evaluar la asociación entre la microbiota intratumoral y la respuesta al tratamiento en cáncer colorrectal, considerando las diferencias según sexo, mediante un análisis secundario de datos metagenómicos de la cohorte TCGA-COAD-READ integrados con el repositorio The Cancer Microbiome Atlas (TCMA). Se analizaron 90 muestras tumorales primarias. La microbiota intratumoral estuvo dominada principalmente por los filos Bacteroidetes, Firmicutes y Proteobacteria. No se observaron diferencias estadísticamente significativas en la abundancia relativa de los principales filos bacterianos ni en los índices de diversidad alfa entre pacientes respondedores y no respondedores, ni entre hombres y mujeres. Sin embargo, se observaron diferencias descriptivas en la abundancia relativa de algunos filos según sexo y respuesta terapéutica. En conclusión, en esta cohorte no se identificaron asociaciones estadísticamente significativas entre la microbiota intratumoral, la respuesta al tratamiento y el sexo. Estos resultados sugieren la necesidad de estudios con mayor tamaño muestral y enfoques funcionales que permitan comprender mejor el papel de la microbiota intratumoral en el cáncer colorrectal.

Colorectal cancer (CRC) is one of the leading causes of morbidity and mortality worldwide. In recent years, the intratumoral microbiota has been proposed as a relevant factor in tumor progression and treatment response. Additionally, biological sex may influence microbiome composition and function, highlighting the importance of studying these interactions. The aim of this study was to evaluate the association between intratumoral microbiota and treatment response in colorectal cancer, considering sex differences, through a secondary analysis of metagenomic data from the TCGA-COAD-READ cohort integrated with The Cancer Microbiome Atlas (TCMA). A total of 90 primary tumor samples were analyzed. The intratumoral microbiota was mainly dominated by Bacteroidetes, Firmicutes, and Proteobacteria. No statistically significant differences were observed in the relative abundance of major bacterial phyla or in alpha diversity indices between responders and non-responders, nor between males and females. However, descriptive differences in relative abundance were observed across groups. In conclusion, no statistically significant associations were identified between intratumoral microbiota, treatment response, and sex in this cohort. These findings highlight the need for larger studies and functional approaches to better understand the role of the microbiota in colorectal cancer.
