La neurocisticercosis (NCC) es una enfermedad parasitaria del sistema nervioso central asociada a una respuesta inflamatoria que contribuye al daño cerebral y a manifestaciones neurológicas como epilepsia y deterioro cognitivo. La piroptosis es un tipo de muerte celular inflamatoria regulada que ha sido implicada en diversas enfermedades neuroinflamatorias; sin embargo, su participación en la neurocisticercosis no ha sido ampliamente estudiada. El objetivo del presente estudio fue evaluar la inmunorreactividad de biomarcadores asociados a la piroptosis, inflamasoma NLRP3, caspasa-1, interleucina-1β (IL-1β) y la forma clivada de gasdermina D (GSDMD-C1), en tejido cerebral de ratas con neurocisticercosis, en comparación con ratas no infectadas. Se utilizaron cortes histológicos de cerebro fijados en parafina, previamente obtenidos de un estudio experimental, los cuales fueron analizados mediante inmunohistoquímica. Asimismo, se realizó inmunofluorescencia de doble marcaje para identificar la población celular inmunorreactivas a GSDMD-C1. La cuantificación se realizó mediante un análisis semi-cuantitativo de área inmunorreactiva y se aplicaron pruebas estadísticas no paramétricas. Los resultados mostraron una mayor inmunorreactividad de NLRP3, caspasa-1, GSDMD-C1 e IL-1β en los tejidos cerebrales de ratas infectadas, con variaciones dependientes del tiempo posinfección. GSDMD-C1 fue el biomarcador que presentó un aumento más consistente en los diferentes tiempos evaluados. El análisis de correlación evidenció asociaciones moderadas entre NLRP3 y GSDMD-C1, así como entre caspasa-1 e IL-1β. La inmunofluorescencia mostró expresión de GSDMD-C1 en neuronas y astrocitos. En conjunto, estos hallazgos sugieren que la neurocisticercosis induce la activación de componentes clave de la vía piroptótica en el tejido cerebral, contribuyendo a la respuesta inflamatoria asociada a esta enfermedad.

Neurocysticercosis (NCC) is a parasitic disease of the central nervous system associated with an inflammatory response that contributes to brain damage and neurological manifestations such as epilepsy and cognitive impairment. Pyroptosis is a type of regulated inflammatory cell death that has been implicated in various neuroinflammatory diseases; however, its role in neurocysticercosis has not been extensively studied. The objective of this study was to evaluate the immunoreactivity of biomarkers associated with pyroptosis, imflamosome NLRP3, caspase-1, interleukin 1β (IL-1β), and the cleaved form of gasdermin D (GSDMD C1), in brain tissue from rats with neurocysticercosis, compared to healthy rats. Paraffin-embedded brain histological sections, previously obtained from an experimental study, were analyzed using immunohistochemistry. Additionally, double-labeling immunofluorescence was performed to identify cell populations immunoreactive to GSDMD C1. Quantification was performed using a semi-quantitative analysis of immunoreactive area, and nonparametric statistical tests were applied. The results showed increased immunoreactivity of NLRP3, caspase-1, GSDMD-C1, and IL-1β in the brain tissues of infected rats, with variations depending on the time post-infection. GSDMD-C1 was the biomarker that showed the most consistent increase across the different time points evaluated. Correlation analysis revealed moderate associations between NLRP3 and GSDMD-C1, as well as between caspase-1 and IL-1β. Immunofluorescence showed expression of GSDMD-C1 in ne urons and astrocytes. Taken together, these findings suggest that neurocysticercosis induces the activation of key components of the pyroptotic pathway in brain tissue, contributing to the inflammatory response associated with this disease.
