Dentro de la biología moderna, el modelamiento de la estructura tridimensional de las proteínas proporciona información relevante e irremplazable para determinar su función o características. Uno de los métodos más usados es la cristalografía de rayos X; para ello es necesario obtener cristales de proteína. Una de las técnicas más usadas es la difusión por vapor; en la cual una gota de muestra pura y homogénea con precipitante llega a la sobresaturación. Se pueden obtener diferentes resultados aparte de cristales, como la formación de Spherulites; que son gotas de proteína concentrada y uniforme. Las Spherulites presentan un buen punto de inicio en la optimización del proceso de formación de cristales proteicos. En el 2011 se obtuvo un primer cristal y posterior estructura de la enzima pirazinamidasa de Mycobacterium tuberculosis de la cepa H37Rv.Mediante el método de gota colgante, que usa el principio de la difusión de vapor, y MgSO4 como precipitante; sin embargo, nunca se pudo reproducir el resultado con las condiciones determinadas en dicha investigación. En este estudio mediante el método de gota colgante se determinó cuáles son las mejores condiciones para obtener un estado de Spherulite de la pirazinamidasa mutante K48T. Las Spherulites de pirazinamidasa K48T fueron obtenidas a 1.5M de MgSO4, 100 mM de BME, 5, 10 y 20mg/ml de PZAsa a pH 6.5 de buffer Bis TRIS a 25°C.
Within modern biology, the modeling of the three-dimensional structure of proteins provides relevant and irreplaceable information to determine their function or characteristics. One of the most used methods is X-ray crystallography; for this it is necessary to obtain protein crystals; by methods such as vapor diffusion. In which a drop of pure and homogeneous sample with precipitant reaches supersaturation and changes are observed in the drop. Different results can be obtained apart from crystals, such as the formation of Spherulites, concentrated and uniform protein droplets, which present a good starting point in optimizing the process of protein crystal formation. In 2011, a first crystal and subsequent structure of the Mycobacterium tuberculosis pyrazinamidase enzyme of the H37Rv strain was obtained, using the hanging drop method and MgSO4 as precipitant; however, the result could never be reproduced with the conditions determined in that investigation. In this study using the hanging drop method will be extended are the best conditions to obtain a Spherulite state of the K48T mutant pyrazinamidase. Pyrazinamidase K48T Spherulites were obtained in 1.5M MgSO4, 100 mM BME, 5, 10 and 20mg/ml PZAsa in pH 6.5 Bis TRIS buffer at 25°C.
