Antecedentes: La artritis reumatoide (AR) es una enfermedad autoinmune que causa daño articular y discapacidad. La Persea caerulea, planta utilizada en medicina tradicional peruana, podría tener efectos antiinflamatorios debido a sus flavonoides. Objetivos: Evaluar la citotoxicidad del extracto hidroalcohólico de P. caerulea y su actividad antiinflamatoria en ratas Lewis con AR. Materiales y métodos: Se determinó la citotoxicidad de P. caerulea en células 3T3, Vero, DU145 y SH-SY5Y mediante su IC50. Treinta ratas Lewis se dividieron en seis grupos, cinco con AR inducida y tres de control. Tres grupos recibieron P. caerulea (250, 500 y 1000 mg/Kg) durante 42 días. Se evaluaron con scores de inflamación y grosor de patas. La histología y niveles de TNF-alfa se examinaron post-mortem. Resultados: Los IC50 promedio fueron 60, 130, 194 y 20 μg/mL en 3T3, VERO, DU145 y SH-SY5Y, respectivamente. Los grupos tratados con 500 y 1000 mg/kg de P. caerulea presentaron mayor grosor articular (p<0.05), sin cambios significativos en el score de inflamación. Hubo más inflamación sinovial en ratas con 250 y 500 mg/kg de P. caerulea y más erosión ósea en ratas con 500 y 1000 mg/kg (p<0.05) en comparación con el grupo placebo. Conclusión: El extracto a ≤ 250 mg/mL no fue citotóxico para las líneas celulares. Mostró efectos antiinflamatorios en la primera semana, pero agrava la enfermedad posterior a esta. Se necesitan más estudios para evaluar su efecto antiinflamatorio a corto plazo. |

Background: Rheumatoid arthritis (RA) is an autoimmune disease that causes joint damage and disability. Persea caerulea, a plant used in traditional Peruvian medicine, may have anti-inflammatory effects due to its flavonoids. Objectives: To evaluate the cytotoxicity of the hydroalcoholic extract of P. caerulea and its anti-inflammatory activity in Lewis rats with RA. Materials and Methods: The cytotoxicity of P. caerulea was determined on 3T3, Vero, DU145, and SH-SY5Y cells by its IC50. Thirty Lewis rats were divided into six groups, five with induced RA and three controls. Three groups received P. caerulea (250, 500, and 1000 mg/kg) for 42 days. They were evaluated with inflammation scores and leg thickness. Histology and TNF-alpha levels were examined post-mortem. Results: The average IC50 values were 60, 130, 194, and 20 μg/mL in 3T3, VERO, DU145, and SH-SY5Y, respectively. Groups treated with 500 and 1000 mg/kg of P. caerulea showed increased joint thickness (p<0.05), with no significant changes in the inflammation score. There was more synovial inflammation in rats with 250 and 500 mg/kg of P. caerulea and more bone erosion in rats with 500 and 1000 mg/kg (p<0.05) compared to the placebo group. Conclusion: The extract at ≤ 250 mg/mL was not cytotoxic to the cell lines. It showed anti-inflammatory effects in the first week but aggravated the disease thereafter. Further studies are needed to evaluate its short-term anti-inflammatory effect. |
