El objetivo de esta revisión fue sintetizar de forma sistemática los artículos relacionados a la respuesta inmune frente al virus de la rabia, los mecanismos de evasión utilizados por el mismo, y revisar las perspectivas de vacunación antirrábica en fauna silvestre. La revisión se llevó a cabo usando las bases de datos PUBMED, SCIENCE DIRECT y EUROPE PMC. Cincuenta y tres estudios publicados entre 2004 – 2024 reportan mecanismos de respuesta inmune frente al virus de la rabia en modelos animales. La respuesta inmune innata está representada principalmente por la función de TLR4 y TLR7, así como la sobrerregulación de IFIT3, IFIT2, IIGP1, TNF – α, RIG – I y NLRP3. La respuesta inmune adaptativa está representada únicamente por infiltración de células B, células dendríticas y linfocitos T de memoria residentes. Los mecanismos de evasión inmunológica más resaltantes se tiene el antagonismo de IFN – γ por el bloqueo de las vías NF – κB y JAK – STAT por la interacción de las proteínas virales. Por otro lado, ocho artículos publicados los últimos 20 años reportan datos de la aplicación de vacunas orales en fauna silvestre en diversos países, así como los resultados obtenidos. El caso de Serbia y su vacuna oral en zorros demostró alta eficiencia en el control y prevención de la rabia. Actualmente se tiene como objetivo inmunizar las poblaciones de murciélagos con vacunas oro nasales, tópicas y auto diseminadas con el avance de la ingeniería genética y la inmunoproteómica. Se requieren más estudios de inmunoprevención en fauna silvestre bajo un enfoque “one health”.The aim of this review was to systematically synthesize the articles related to the immune response against the rabies virus, the evasion mechanisms used by it, and to review the perspectives of anti-rabies vaccination in wildlife. The review was carried out using the PUBMED, SCIENCE DIRECT and EUROPE PMC databases. Fifty-three studies published between 2004 and 2024 report immune response mechanisms against the rabies virus in animal models. The innate immune response is represented mainly by the function of TLR4 and TLR7, as well as the upregulation of IFIT3, IFIT2, IIGP1, TNF – α, RIG – I and NLRP3. The adaptive immune response is represented only by infiltration of B cells, dendritic cells and resident memory T lymphocytes. The most notable immune evasion mechanisms are the antagonism of IFN-γ by blocking the NF-κB and JAK-STAT pathways through the interaction of viral proteins. On the other hand, eight articles published in the last 20 years report data on the application of oral vaccines in wildlife in various countries as well as the results obtained. The case of Serbia and its oral vaccine in foxes demonstrated high efficiency in the control and prevention of rabies. Currently, the objective is to immunize bat populations with oronasal, topical and self-disseminated vaccines with the advances of genetic engineering and immunoproteomics. More studies of immunoprevention in wildlife are required under a “one health” approach.
