El siguiente trabajo tiene como objetivo validar una metodología para la cuantificación de los ácidos grasos trans en aceites vegetales mediante la cromatografía de gases, esto con la finalidad de brindar un nuevo servicio para una clientela interesada en obtener la concentración de estos tipos de ácidos grasos en muestras de aceites vegetales. El procedimiento de validación garantiza la confiabilidad de los resultados otorgados por el laboratorio Alex Stewart, para lo cual, se propuso evaluar cuatro métodos analíticos en términos de porcentaje de recuperación, de los cuales se seleccionó el más adecuado en función a sus tiempos de elución, resolución de los picos y la relación señal/ruido. Para validar se utilizó dos estándares de ácidos grasos, Nu-Chek 67 y Nu-Chek 607 junto a un patrón interno (muestra interlaboratorio). Para los ácidos grasos, los límites de detección (LD) fueron, Miristelaidato de metilo - 6.03 ppm, Palmitelaidato de metilo - 6.28 ppm, Petroelaidato de metilo - 5.76 ppm, Elaidato de metilo - 7.09 ppm, Transvacenato de metilo - 9.17 ppm, Linoelaidato de metilo -5.68 ppm, mientras que sus límites de cuantificación (LQ) fueron, Miristelaidato de metilo - 18.27 ppm, Palmitelaidato de metilo - 19.03 ppm, Petroelaidato de metilo - 17.46 ppm, Elaidato de metilo - 21.50 ppm, Transvacenato de metilo - 27.79 ppm y Linoelaidato de metilo - 17.22 ppm. Para evaluar la exactitud y precisión se usó la sumatoria de las áreas de integración de los ácidos grasos C18:1 (ácidos grasos de 18 C con 1 insaturación) y C18:2 (ácidos grasos de 18 C con 2 insaturaciones) cuyos valores son de 21.08% y 2.79% respectivamente. Estos valores se encuentran dentro de las tolerancias propuestas por el certificado. Por otro lado, la precisión medida para C18:1 y C18:2 se calculó como porcentaje de desviación estándar relativa (%RSD), situándose entre 1.78% y 5.52%.
The following work aims to validate a methodology for the quantification of trans fatty acids in vegetable oils by gas chromatography, with the goal of providing a new service for a clientele interested in obtaining the concentration of these types of fatty acids in vegetable oil samples. The validation procedure guarantees the reliability of the results provided by the Alex Stewart laboratory, for which it was proposed to evaluate four analytical methods in terms of recovery percentage and from which the most adequate was selected according to their elution times, peak resolution and signal/noise ratio. For fatty acids, the limits of detection (LOD) obtained were, Methyl myristelaidate - 6.03 ppm, Methyl palmitelaidate - 6.28 ppm, Methyl petroelaidate - 5.76 ppm, Methyl elaidate - 7.09 ppm, Methyl transvacenate - 9.17 ppm, Methyl linolenidate -5. 68 ppm, while their limits of quantification (LQ) were, Methyl myristelaidate - 18.27 ppm, Methyl palmitelaidate - 19.03 ppm, Methyl petroelaidate - 17.46 ppm, Methyl elaidate - 21.50 ppm, Methyl transvacenate - 27.79 ppm and Methyl linoelaidate - 17.22 ppm. To evaluate the accuracy and precision, the sum of the integration areas of the fatty acids C18:1 (18 C fatty acids with 1 unsaturation) and C18:2 (18 C fatty acids with 2 unsaturations) whose values are 21.08% and 2.79%, respectively, were used. These values are within the tolerances proposed by the certificate. On the other hand, the precision measured for C18:1 and C18:2 was calculated as percent relative standard deviation (%RSD), ranging from 1.78% to 5.52%.
