La resistencia antibiótica en Mycobacterium tuberculosis (Mtb) es una preocupación global. La eficacia de los antibióticos dirigidos a la proteína ARN polimerasa (ARNP) la convierte en un blanco apropiado para la creación de nuevos agentes antituberculosos. Sin embargo, el cribado de antibióticos para Mtb conlleva desafíos y riesgos adicionales. Recientemente, el Laboratorio de Moléculas Individuales de la UPCH diseñó una E. coli que expresa una proteína fluorescente verde (GFP), dependiente de la actividad de la ARNP de Mtb producida en su interior. Esta Bacteria Reportera permitiría el cribado de nuevos inhibidores de esta enzima, reduciendo así los riesgos para la salud, gastos y tiempo. Sin embargo, los posibles efectos en E. coli podrían dificultar la interpretación de resultados. Por ello, hemos planteado evaluar antibióticos con acción sobre E. coli para discernir entre efectos sobre la ARNP de Mtb y sobre la célula hospedera. Se seleccionó un antibiótico que afecta la viabilidad de E. coli (Cefuroxima) y otro que afecta a sus ribosomas (Cloranfenicol). Se evaluaron sus efectos sobre la supervivencia bacteriana, la fluorescencia normalizada, así como las tasas de crecimiento y fluorescencia de GFP. Además, se comparó su acción con otros antibióticos que inhiben la actividad de ARNP-Mtb (Rifaximina y Rifampicina). Para todos los antibióticos se observó una tendencia común: una reducción en la fluorescencia que no necesariamente se correlaciona con una disminución del crecimiento. Esta observación fue respaldada por un modelo matemático que mostró un resultado similar, incluso para la Cefuroxima. Sin embargo, los gráficos de fluorescencia indicaron que los antibióticos que afectan a la ARNP-Mtb generan una caída más lenta y abrupta de producción de GFP que aquellos que solo afectan a la bacteria hospedera. Aunque el sistema actual puede usarse para el cribado de antibióticos dirigidos a la ARNP-Mtb, es necesario evaluar a otros antibióticos con diferentes blancos de acción.
Antibiotic resistance in Mycobacterium tuberculosis (Mtb) is a global concern. The efficacy of antibiotics targeting the RNA polymerase protein (ARNP) makes it an appropriate target for the development of novel anti-tuberculosis agents. However, antibiotic screening for Mtb carries additional challenges and risks. Recently, the Laboratorio de Moléculas Individuales at UPCH engineered an E. coli that expresses a green fluorescent protein (GFP), dependent on the Mtb ARNP activity produced within it. This reporter bacterium would allow the screening of new inhibitors of this enzyme, thus reducing health risks, expenses and time. However, possible effects on E. coli could make interpretation of results difficult. Therefore, we set out to evaluate antibiotics with action on E. coli to discern between effects on Mtb ARNP and on the host cell. An antibiotic that affects the viability of E. coli (Cefuroxime) and another that affects its ribosomes (Chloramphenicol) were selected. Their effects on bacterial survival, normalized fluorescence, as well as growth and GFP fluorescence rates were evaluated. In addition, their action was compared with other antibiotics that inhibit ARNP-Mtb activity (Rifaximin and Rifampicin). For all antibiotics, a common trend was observed: a reduction in fluorescence that does not necessarily correlate with a decrease in growth. This observation was supported by a mathematical model that showed a similar result, even for Cefuroxime. However, the fluorescence plots indicated that antibiotics that affect ARNP-Mtb generate a slower and steeper drop in GFP production than those that only affect the host bacterium. Although the current system can be used for screening antibiotics targeting ARNP-Mtb, other antibiotics with different targets of action need to be evaluated.
