El cáncer es una enfermedad que afecta tanto a humanos como animales domésticos y silvestres. Cada célula y año de vida adicional deberían aumentar la probabilidad de carcinogénesis, sin embargo, la tasa de ocurrencia de cáncer no está en función directa con el tamaño del animal. La paradoja de Peto señala que no hay asociación entre el tamaño corporal, la longevidad y el riesgo de cáncer entre las especies y predice que los animales con un gran tamaño corporal desarrollarían mecanismos únicos para evadir el cáncer. Se han planteado distintas hipótesis para resolverla y si bien entre ellas se ha mencionado a los genes de reparación del ADN, estos no han sido investigados de manera específica y a un nivel estructural y funcional de sus proteínas. La reparación del ADN es un mecanismo que evita la acumulación de mutaciones en el genoma y sus vías son altamente conservadas. En la carcinogénesis, la maquinaria de reparación del ADN está dañada, esto genera inestabilidad genómica que promueve la transformación celular. La unión de las proteínas reparadoras de ADN al ácido nucleico se da por medio de dominios que muestran una alta conservación de residuos. Se ha documentado que las ballenas muestran una estrategia de evasión del cáncer inclinada a la reparación del ADN puesto que presentan amplificaciones en sus genes reparadores, mientras que, los elefantes poseen una estrategia dirigida hacia la apoptosis por medio de TP53. Esto sugiere que las ballenas tienen un mecanismo de reparación del ADN más eficiente y exitoso en comparación con los elefantes, lo cual podría tener su explicación en la estructura y función de las proteínas reparadoras. Se plantea un estudio comparativo de las secuencias aminoacídicas y estructuras 3D de las proteínas reparadoras de ADN de humanos ortólogas en ballenas, elefantes y ratones.

Each additional cell and year of life should increase the probability of carcinogenesis, nevertheless, the rate of cancer does not have a direct function with the size of the animal. The Peto paradox points out that there is no association between body size, longevity, and cancer risk between species and predicts that animals with large body sizes would develop unique mechanisms to evade cancer. Different hypotheses have been put forward to solve it, and although DNA repair genes have been mentioned among them, they have not been specifically investigated at a structural and functional level of their proteins. DNA repair is a mechanism that prevents the accumulation of mutations in the genome and its pathways are highly conserved. In carcinogenesis, the DNA repair machinery is damaged, this generates genomic instability that promotes cell transformation. The binding of DNA repair proteins to nucleic acid is through domains that show high residue conservation. It has been documented that whales show a cancer avoidance strategy inclined to DNA repair since they present amplifications in their repair genes, whereas elephants have a strategy directed towards apoptosis through TP53. This suggests that whales have a more efficient and successful DNA repair mechanism compared to elephants, which could be explained by the structure and function of repair proteins. A comparative study of the amino acid sequences and 3D structures of orthologous human DNA repair proteins in whales, elephants and mice is proposed.
