La infección pulmonar por Mycobacterium tuberculosis se asocia con el cáncer de pulmón. Así, las personas con historial de tuberculosis pulmonar tienen más riesgo de desarrollar cáncer de pulmón que aquellas que no tuvieron tuberculosis. Sin embargo, existe poca información sobre la participación de M. tuberculosis en la etiología de la neoplasia pulmonar. Se ha descrito que M. tuberculosis daña el ADN de las células humanas, aunque no se sabe cómo. Un posible mecanismo sería a través de proteínas secretadas como parte de su virulencia, que se dirijan al núcleo. Por ello, en este estudio nos propusimos predecir las proteínas secretadas por M. tuberculosis que se dirigen al núcleo de la célula huésped, mediante una difusión pasiva, y su potencial interacción con el ADN. Utilizando programas bioinformáticos, descubrimos que de las 972 proteínas que componen el secretoma de M. tuberculosis, 58 proteínas tienen como destino el núcleo de las células de su hospedador. Así mismo, de ellas, encontramos que 3 proteínas (la Proteína ribosómica 30S S3, la proteína reguladora transcripcional Rv3488 y la Formamidopirimidina-ADN glicosilasa) poseen una alta probabilidad de interactuar directamente con el ADN. Nuestros resultados respaldarían la hipótesis que vincula la infección por M. tuberculosis y el cáncer de pulmón, pudiendo tener, este grupo de proteínas de secreción destinadas al núcleo, potencial para alterar las funciones del genoma al interactuar con los ácidos nucleicos.
Lung infection with Mycobacterium tuberculosis is associated with lung cancer. Thus, individuals with a history of pulmonary tuberculosis are at higher risk of developing lung cancer. However, there is little information on the involvement of M. tuberculosis in the etiology of lung neoplasia. It has been described that M. tuberculosis damages the DNA of human cells, although it is not known how. One possible mechanism would be through secreted proteins that target the nucleus as part of its virulence. In the present study, we set out to predict the proteins secreted by M. tuberculosis that target the host cell nucleus via passive diffusion, and their potential interaction with DNA. Using bioinformatics procedures, we discovered that of the 972 proteins that make up the M. tuberculosis secretome, 58 proteins target the host cell nucleus. Of these, 3 proteins (30S ribosomal protein S3, transcriptional regulatory protein Rv3488 and formamidopyrimidine-DNA glycosylase) have a high probability of interacting directly with DNA. Our results support the link between M. tuberculosis infection and lung cancer, and this group of secretion proteins destined to the nucleus may have the potential to alter genome functions by interacting with nucleic acids.
